Supplementary information

COMMD7 also known as Copper Metabolism MURR1 Domain-containing protein 7 is a protein with a mass of approximately 22 kDa. This protein participates in the regulation of copper ion homeostasis by influencing the expression of genes involved in copper transport. It is expressed in various tissues including the liver and kidneys where it modulates cellular levels of copper necessary for maintaining metabolic balance. The regulation by COMMD7 affects transcription factors contributing to both the inhibition and activation of certain genes.
Biological function summary

COMMD7 plays a role in controlling inflammation and oxidative stress. It interacts with other COMMD family members forming a complex that influences NF-κB signaling pathways. This pathway is important for immune response regulation making COMMD7 critical for managing immune system activities and processes. By regulating NF-κB COMMD7 also contributes to the cell's response to stress and modulates inflammatory cytokine production impacting systemic inflammation levels.

Pathways

COMMD7 functions in the NF-κB and HIF-1α signaling pathways both important for cellular stress responses. These pathways control transcriptional programs linked to inflammation immunity and adaptive responses to low oxygen environments. COMMD7's interactions with IκB proteins and HIF-1α demonstrate its involvement in pathway regulation wherein it assists in managing the stability and activity of these proteins during cellular adaptation to environmental changes.

The dysregulation of COMMD7 associates with several conditions such as cancer and inflammatory diseases. Its role in these disorders involves interaction with pathways impacting cell proliferation and immune system modulation. Aberrant COMMD7 expression may influence NF-κB activity contributing to chronic inflammation seen in inflammatory diseases or tumor progression in cancers. Studies have shown correlations between COMMD7 IκB proteins and HIF-1α in these diseases highlighting its potential as a therapeutic target.
