Function

Factor B which is part of the alternate pathway of the complement system is cleaved by factor D into 2 fragments: Ba and Bb. Bb, a serine protease, then combines with complement factor 3b to generate the C3 or C5 convertase. It has also been implicated in proliferation and differentiation of preactivated B-lymphocytes, rapid spreading of peripheral blood monocytes, stimulation of lymphocyte blastogenesis and lysis of erythrocytes. Ba inhibits the proliferation of preactivated B-lymphocytes.

Activity summary

Complement Factor B also known as CFB Ba fragment or C3 proactivator participates mechanically in the alternative complement pathway. It has a molecular mass of approximately 93 kDa. Expressed mainly in the liver and found in plasma Factor B cleaves into two fragments Bb and Ba by Factor D. The Ba fragment specifically is known for its interactions that modulate complement activation. Ba itself although smaller is an active part of this cleavage event.

Biological function summary

Complement Factor B is important in the innate immune response. It forms a complex called the C3 convertase when combined with C3b and Bb fragment which amplifies the cascade leading to pathogen opsonization and lysis. The Ba fragment though not part of the convertase influences cell signaling and modulates inflammatory responses. It directly affects immune cells by binding to receptors and therefore impacts the regulation of immune responses.

Pathways

The alternative complement pathway and the regulation of inflammation fall under the influence of Complement Factor B. Within the complement pathway its interaction with proteins like C3 and Factor D emphasizes its position. This pathway is part of wider immune management processes highlighting its key role interacting with these proteins to control activation and amplification of the complement system leading to enhanced pathogen clearance and aiding the defense mechanism.

Associated diseases and disorders

Complement Factor B shows connections to age-related macular degeneration (AMD) and atypical hemolytic uremic syndrome (aHUS). In AMD abnormal regulation of the complement pathway with involvement of Complement Factor B contributes to drusen formation complicating the disease progression. It also links to aHUS where uncontrolled complement activation causes endothelial damage. Proteins such as Complement Factor H which modulates the alternative pathway show a significant relationship with Factor B in these disease processes affecting the progression and control of these disorders through complement regulation.
