Supplementary information

Connecdenn 3 also known as DENND1C is a guanine nucleotide exchange factor with a mass of approximately 125 kDa. It plays an important role in cell signaling by activating small GTPases. The protein is primarily expressed in human immune cells with significant presence in dendritic cells. It has a part in modulating endocytic trafficking impacting vesicle formation and cargo sorting.
Biological function summary

This protein is part of the DENN domain proteins family known to regulate endosomal functions. Connecdenn 3 participates in the process of endocytosis facilitating the movement of materials into the cell. Through its role as a guanine nucleotide exchange factor it affects the activation of Rab small GTPases which are essential for intracellular transport and recycling of membrane components.

Pathways

Connecdenn 3 functions centrally in endocytic and recycling pathways. It collaborates with Rab proteins important for cargo selection and vesicle budding. Through these interactions it influences the membrane trafficking machinery and cellular signaling networks connected to the movement of vesicles. It has a relationship with the Ras-related protein Rab-5 impacting pathways associated with the transport and sorting of endocytic vesicles.

Connecdenn 3 links to immunological conditions and is associated with allergies. Altered expression levels or mutations in Connecdenn 3 can disrupt Rab GTPase activity affecting immune responses. The protein's relationship with Rab-5 suggests its involvement in disorders where vesicle trafficking and cellular communication are impaired potentially contributing to abnormal immune function or inflammatory processes.

Target data

Guanine nucleotide exchange factor (GEF) which may activate RAB8A, RAB13 and RAB35. Promotes the exchange of GDP to GTP, converting inactive GDP-bound Rab proteins into their active GTP-bound form.
