Contactins mediate cell surface interactions during nervous system development. Involved in the formation of paranodal axo-glial junctions in myelinated peripheral nerves and in the signaling between axons and myelinating glial cells via its association with CNTNAP1. Participates in oligodendrocytes generation by acting as a ligand of NOTCH1. Its association with NOTCH1 promotes NOTCH1 activation through the released notch intracellular domain (NICD) and subsequent translocation to the nucleus. Interaction with TNR induces a repulsion of neurons and an inhibition of neurite outgrowth (By similarity).

Contactin 1 also known as contactin-1 or contactin is a protein involved in cell adhesion processes. It is sometimes referred to as contacton. This protein has a molecular weight of approximately 120 kDa and is a neural membrane glycoprotein. Contactin 1 primarily expresses in the developing nervous system and persists in the adult central nervous system. It plays an important role in forming connections between neurons and ensuring proper communication between them.

Biological function summary

The role of this protein involves maintaining neuronal network organization and integrity. Contactin 1 plays a critical part in myelination a process essential for the swift conduction of nerve impulses. It interacts with several partners forming complexes essential for neuronal stability including neurofascin and contactin-associated protein-like 2. These interactions facilitate proper signal transmission across synapses in the nervous system.

Contactin 1 is an important element in the pathways that regulate neuronal development and myelination. It is involved in the formation of the nodes of Ranvier important for nerve impulse transmission.
