Supplementary information

COX10 also known as Protoheme IX farnesyltransferase plays a significant role in the heme A biosynthesis pathway which is essential for the maturation of cytochrome c oxidase (Complex IV). The molecular mass of COX10 is approximately 48 kDa. It is an inner mitochondrial membrane protein primarily expressed in tissues with high metabolic rates such as muscles and the liver. COX10 facilitates the conversion of protoheme IX into heme O by transferring a farnesyl group a critical step in heme A production.
Biological function summary

COX10 functions as part of the cytochrome c oxidase complex serving an important role in cellular respiration. This enzyme complex regulates the final step of the mitochondrial electron transport chain which is vital for ATP production. Heme A synthesized with the help of COX10 is necessary for the proper function of cytochrome c oxidase which catalyzes the reduction of oxygen to water. Therefore COX10 indirectly contributes to the maintenance of cellular energy homeostasis.

Pathways

COX10 is integral to the oxidative phosphorylation pathway a central route in cellular metabolism. Through its involvement in this pathway COX10 affects the electron transport chain and directly impacts ATP synthesis. COX10 interacts with other cytochrome c oxidase subunits such as COX15 which also participates in heme A biosynthesis. These interactions emphasize the role of COX10 in coordination with other proteins to support efficient energy production.

Dysfunction in COX10 can contribute to disorders like mitochondrial complex IV deficiency and Leigh syndrome a severe neurological disorder. Mutations in the COX10 gene can disrupt cytochrome c oxidase assembly or activity leading to reduced ATP production and ensuing energy deficits in cells. This dysfunctional energy metabolism impacts tissues with high energy demands linking COX10 to disease pathology. Additionally COX15 and its role in heme A biosynthesis may further elucidate the disease mechanisms associated with COX10-related disorders.
