Immunohistochemical analysis of paraffin-embedded human pancreas tissue labeling CPE with ab275882 at 1/2000 (0.266 μg/ml) dilution followed by a ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101). Cytoplasmic staining on human pancreas islets (PMID : 21152387). The section was incubated with ab275882 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.

Secondary antibody only control : Secondary antibody is a ready to use Rabbit specific IHC polymer detection kit HRP/DAB (ab209101).

Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.

Immunohistochemical analysis of 4% PFA-fixed, 0.2% Triton X-100 permeabilized frozen mouse pancreas tissue labeling CPE with ab275882 at 1/100 (5.31 μg/ml) dilution followed by ab150077 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (Green). Positive staining on mouse pancreatic islet is observed. The nuclear counterstain was DAPI (Blue).

Carboxypeptidase E (CPE) also known as carboxypeptidase H is an enzyme with a mass of about 53 kDa. It facilitates the removal of C-terminal amino acid residues from proteins and peptides. CPE is primarily expressed in endocrine and neuroendocrine tissues including the brain pituitary and pancreas. Its expression in these areas suggests significant involvement in the processing and regulation of peptide hormones and neurotransmitters.
Biological function summary

CPE influences the maturation of peptide hormones and neurotransmitters converting inactive precursors into active forms. CPE does not function as part of a larger complex but acts independently to cleave the C-terminal residues. Its activity plays a role in the biosynthesis of hormones like insulin and proinsulin regulating energy homeostasis and metabolic processes.

Pathways

The activities of enzymes like CPE are integral to the prohormone and neuropeptide processing pathways. Within these pathways CPE works alongside other processing enzymes such as prohormone convertases to ensure proper hormone maturation and secretion. In these pathways CPE connects with proteins like chromogranin A which assists in the storage and trafficking of secretory granules in neuroendocrine cells.

Dysregulation of CPE has links to obesity and type 2 diabetes due to its role in peptide hormone processing. Altered CPE function can impair the maturation of insulin and other hormones contributing to these metabolic conditions. In addition relationships between CPE and other proteins such as proinsulin and insulin further highlight its significance in the pathology of these diseases.
