Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

The target CPNE1 also known as Copine-1 is a calcium-dependent phospholipid-binding protein. It weighs approximately 59 kDa and is expressed in various tissues including the brain heart and skeletal muscle. CPNE1 has eight conserved regions which include two C2 domains and a von Willebrand factor A domain enabling its function in the cell.
Biological function summary

CPNE1 interacts with membrane lipids and proteins playing a significant role in diverse cellular processes. It is part of a larger complex associated with membrane trafficking and cytoskeletal organization. CPNE1 can influence cellular processes like cell division and motility through its interactions with other proteins often regulating intracellular signaling pathways.

Pathways

CPNE1 is involved in calcium-mediated signaling and the phosphoinositide 3-kinase (PI3K) pathway. The protein interacts with Akt a downstream target of the PI3K pathway therefore influencing cell survival and growth. These interactions underline CPNE1's role in essential cellular functions linking it with other proteins like phospholipase C.

CPNE1 has associations with cancer progression and neurodegenerative diseases. It interacts with proteins like p53 which play important roles in cell cycle regulation and apoptosis. In cancer altered CPNE1 activity can disrupt normal cell signaling potentially leading to uncontrolled cell growth. In neurodegenerative diseases its involvement in cell signaling pathways may contribute to neuronal dysfunction or death.

Target data

Calcium-dependent phospholipid-binding protein that plays a role in calcium-mediated intracellular processes (PubMed : 14674885). Involved in the TNF-alpha receptor signaling pathway in a calcium-dependent manner (PubMed : 14674885). Exhibits calcium-dependent phospholipid binding properties (PubMed : 19539605, PubMed : 9430674). Plays a role in neuronal progenitor cell differentiation; induces neurite outgrowth via a AKT-dependent signaling cascade and calcium-independent manner (PubMed : 23263657, PubMed : 25450385). May recruit target proteins to the cell membrane in a calcium-dependent manner (PubMed : 12522145). May function in membrane trafficking (PubMed : 9430674). Involved in TNF-alpha-induced NF-kappa-B transcriptional repression by inducing endoprotease processing of the transcription factor NF-kappa-B p65/RELA subunit (PubMed : 18212740). Also induces endoprotease processing of NF-kappa-B p50/NFKB1, p52/NFKB2, RELB and REL (PubMed : 18212740).
