CRB2 also known as Crumbs homolog 2 is a protein involved in important cellular functions. It is part of the Crumbs family and has a molecular weight of approximately 137 kDa. Researchers find CRB2 expression largely in the brain and kidneys playing roles in maintaining cell junction integrity. The protein localizes mainly to tight junctions where it plays a mechanically important role in stabilizing cell polarity and tissue organization.
Biological function summary

CRB2 contributes to processes of epithelial cell polarization and morphogenesis. As part of a protein complex involving CRB1 CRB2 and CRB3 it participates in maintaining cell cortex stability. Through this action CRB2 ensures that the cellular architecture remains intact which is essential for proper organ function and development. It influences the formation of apical-basal polarity in epithelial cells which is essential for constructing specialized tissues.

Pathways

CRB2 integrates into the Hippo signaling pathway and tight junction pathway. In the Hippo pathway it closely interacts with proteins like YAP and TAZ to regulate organ size by controlling cell proliferation and apoptosis. By affecting tight junctions CRB2 also influences pathways involved in cell adhesion and the maintenance of cellular barriers connecting with partners such as ZO-1.

CRB2 associates with retinal degeneration and nephrotic syndrome. Mutations affecting its function can lead to retinopathies as its role in maintaining retinal cell structure is compromised. Similarly CRB2's interaction with podocyte proteins is critical in kidney function linking it to nephrotic syndrome when dysfunctional. Pathological conditions also show connections to CRB1 where mutations in both proteins may combine to exacerbate disease states.

Apical polarity protein that plays a central role during the epithelial-to-mesenchymal transition (EMT) at gastrulation, when newly specified mesodermal cells move inside the embryo (By similarity). Acts by promoting cell ingression, the process by which cells leave the epithelial epiblast and move inside the embryo to form a new tissue layer (By similarity). The anisotropic distribution of CRB2 and MYH10/myosin-IIB at cell edges define which cells will ingress : cells with high apical CRB2 are probably extruded from the epiblast by neighboring cells with high levels of apical MYH10/myosin-IIB (By similarity). Plays a role in the maintenance of retinal neuroepithelium organization, structural integrity, adhesion, photoreceptor polarity and retinal photoreceptor layer thickness (By similarity). May play a role in determining the length of cone photoreceptor outer segments and proliferation of late-born progenitor cells (By similarity). Also required for maintenance of the apical polarity complex during development of the cortex (By similarity). Inhibits gamma-secretase-dependent cleavage of APP and secretion of amyloid-beta peptide 40 and amyloid-beta peptide 42, and thereby inhibits gamma-secretase-dependent Notch transcription (PubMed : 20299451).
