Function

Substrate recognition component of a DCX (DDB1-CUL4-X-box) E3 protein ligase complex that mediates the ubiquitination and subsequent proteasomal degradation of target proteins, such as MEIS2, ILF2 or GLUL (PubMed:26990986, PubMed:33009960). Normal degradation of key regulatory proteins is required for normal limb outgrowth and expression of the fibroblast growth factor FGF8 (PubMed:20223979, PubMed:24328678, PubMed:25043012, PubMed:25108355). Maintains presynaptic glutamate release and consequently cognitive functions, such as memory and learning, by negatively regulating large-conductance calcium-activated potassium (BK) channels in excitatory neurons (PubMed:18414909, PubMed:29530986). Likely to function by regulating the assembly and neuronal surface expression of BK channels via its interaction with KCNT1 (PubMed:18414909). May also be involved in regulating anxiety-like behaviors via a BK channel-independent mechanism (By similarity). Plays a negative role in TLR4 signaling by interacting with TRAF6 and ECSIT, leading to inhibition of ECSIT ubiquitination, an important step of the signaling (PubMed:31620128).

Activity summary

CRBN also known as cereblon is a protein involved in the regulation of protein homeostasis. It has a molecular weight of approximately 51 kDa. CRBN is expressed in various tissues including the brain kidney and muscle. It functions as a substrate receptor for the CRL4-CRBN E3 ubiquitin ligase complex recognizing proteins for ubiquitination and degradation. The chemical structure of thalidomide shows binding affinity to CRBN influencing its function.

Biological function summary

Cereblon plays a role in the regulation of protein stability and degradation. It forms part of the CRL4-CRBN complex which tags specific proteins for ubiquitination and subsequent proteasomal degradation. This function is critical for maintaining cellular protein balance. Additionally CRBN modulates immune response and has immunomodulator properties influencing cell-mediated responses.

Pathways

CRBN participates in the ubiquitin-proteasome pathway which regulates protein turnover and quality control within cells. Moreover CRBN is involved in immune signaling pathways partly by modulating the expression of various cytokines and immune-related proteins. It interacts with proteins such as DDB1 and CUL4 within these pathways impacting cellular responses to stress and immune challenges.

Associated diseases and disorders

CRBN is associated with multiple myeloma and intellectual disability. Cereblon acts as a target for drugs like thalidomide and lenalidomide which are used in the treatment of multiple myeloma by altering ubiquitination processes. In intellectual disability alterations in CRBN function or expression can disrupt neuronal development and signaling. CRBN interacts with proteins like IKZF1 and IKZF3 within these contexts affecting their roles in disease progression and treatment responses.

    Lanes 1 - 4: Merged signal (red and green). Green - Anti-CRBN antibody [4D6] ab244223 observed at 55 kDa. Red - loading control, Anti-GAPDH antibody [EPR16891] - Loading Control ab181602 (Rabbit Anti-GAPDH antibody [EPR16891]) observed at 37kDa.

    Anti-CRBN antibody [4D6] ab244223 was shown to react with CRBN in MOLT4 (Licensor sample) wild-type cells in Western blot. Loss of signal was observed when CRBN knockout sample was used. MOLT4 (Licensor sample) wild-type and CRBN knockout cell lysates were subjected to SDS-PAGE. Membranes were blocked in 3% Milk in TBS-T (0.1% Tween®) before incubation with Anti-CRBN antibody [4D6] ab244223 and Anti-GAPDH antibody [EPR16891] - Loading Control ab181602 (Rabbit Anti-GAPDH antibody [EPR16891]) overnight at 4°C at 2 μg/ml and a 1 in 20000 dilution respectively. Blots were developed with Goat anti-Mouse IgG H&L (IRDye® 800CW) preabsorbed (Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed ab216772) and Goat anti-Rabbit IgG H&L (IRDye® 680RD) preabsorbed (Goat Anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed ab216777) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.

    This image was developed using the same antibody clone but in a different formulation, PBS and sodium azide, Anti-CRBN antibody [4D6] ab244223.

    IHC image of CRBN staining in a section of formalin-fixed paraffin-embedded normal human testis* performed on a Leica BOND™ system using the standard Protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20mins. The section was then incubated with Anti-CRBN antibody [4D6] ab244223, 1μg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

    The inset secondary-only control image is taken from an identical assay without primary antibody.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

A band was observed at 51 kDa in wild-type MCF7 cell lysates with no signal observed at this size in CRBN knockout cell line. To generate this image, wild-type and CRBN knockout MCF7 cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Mouse IgG H&L 800CW and Goat anti-Rabbit IgG H&L 680RD at 1/20000 dilution.
