Supplementary information

CREG1 also known as CREG is a glycoprotein that interacts with transcription machinery to regulate cellular growth and morphology. The protein has a molecular mass around 25 kDa. It is expressed in various tissues with the highest expression noted in reproductive organs and the heart. CREG1 acts as a transcriptional regulator by binding to DNA elements and specific proteins such as the retinoblastoma protein (pRb). This binding contributes to its role in modulating gene expression related to cell division and differentiation.
Biological function summary

CREG1 influences cell proliferation and differentiation processes. It forms part of a complex network involving transcription factors and other proteins that govern these cellular functions. Through its interaction with pRb CREG1 can repress certain gene activities while activating others allowing it to maintain a balance between cellular growth and maturation. This balance is essential for normal development and tissue maintenance.

Pathways

CREG1 has a significant role in TGF-beta and E2F-dependent signaling. These pathways are key in cell cycle regulation apoptosis and stem cell biology. Through the E2F pathway CREG1 influences the transcriptional regulation and aids in controlling cell division. CREG1's interaction with pRb and E2F exemplifies how it modulates the progression of the cell cycle playing a part in pathways that determine cell identity and quiescence.

Irregular CREG1 activity links to cancer and cardiac disorders. Specifically abnormal expression levels can lead to cancerous growth through dysregulated cell cycle control partly mediated by its relationship with pRb. In cardiac disorders alterations in CREG1 expression are associated with heart disease as it is involved in cardiac cell homeostasis. These connections highlight the importance of CREG1 in maintaining normal cellular functions in vital organs.
