Biological function summary
Cas12a serves an important role in the CRISPR-Cas adaptive immune system in bacteria. It is part of the Class 2 CRISPR systems and functions in a complex with a single guide RNA (sgRNA). In this complex Cas12a distinguishes itself from other CRISPR-associated proteins by requiring a simpler PAM sequence which offers greater flexibility in target DNA recognition. Cas12a's ability to cleave both strands of DNA with precision makes it a valuable tool for genetic engineering and functional genomics studies.
Pathways
Cas12a enables targeted edits in genetic pathways contributing to advancements in synthetic biology and gene therapy applications. Cas12a interacts with the DNA repair machinery influencing pathways like non-homologous end joining (NHEJ) and homology-directed repair (HDR). These pathways are also influenced by proteins such as DNA ligase IV in NHEJ and RAD51 in HDR showcasing how Cas12a-mediated DNA modifications are repaired and integrated.
