Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Anti-CRISPR protein AcrIIA4 also known simply as AcrIIA4 acts as a potent inhibitor of the CRISPR/Cas9 system. This protein binds to the Cas9 enzyme preventing it from associating with DNA and stopping the gene-editing process. AcrIIA4 has a molecular mass of approximately 12 kDa and is expressed in various bacterial species most notably in Listeria monocytogenes. The mechanical inhibition of the CRISPR activity by AcrIIA4 is specific targeting the Cas9 DNA-binding groove to block its function effectively.
Biological function summary

This anti-CRISPR protein plays a role in bacterial defense against phages that use CRISPR/Cas systems to target bacterial cells. AcrIIA4 operates as part of a protein complex within the bacterial cell working alongside other cell machinery to ensure proper functioning of bacterial genes without interference from CRISPR systems. By blocking the CRISPR/Cas mechanism AcrIIA4 can maintain bacterial genome stability in environments where genetic modification pressure is high.

Pathways

AcrIIA4 participates in bacterial immune evasion pathways notably interfering with the CRISPR-associated immune pathway. The most important pathway related to this is the Type II CRISPR/Cas system where AcrIIA4 directly targets the Cas9 protein. This interaction prevents the cleavage of target DNA by the Cas9 complex offering insight into regulatory mechanisms that control gene-editing processes in both natural and synthetic settings. AcrIIA4 connects with other anti-CRISPR proteins which together modulate the overall impact of CRISPR activity in microbial communities.

AcrIIA4's primary implications relate to bacterial pathogenicity and potential impacts on antimicrobial resistance. Understanding AcrIIA4 in pathogens like Listeria monocytogenes helps unravel its role in microbial resistance to phage therapies which might be used to treat bacterial infections. Additionally as the inhibition mechanism can affect gene-editing efficiency AcrIIA4 is relevant in discussions about potential off-target impacts in CRISPR therapeutic applications mediated through Cas9 misregulation.
