Chromosome cohesion factor involved in sister chromatid cohesion and fidelity of chromosome transmission. Component of one of the cell nuclear antigen loader complexes, CTF18-replication factor C (CTF18-RFC), which consists of CTF18, CTF8, DCC1, RFC2, RFC3, RFC4 and RFC5. The CTF18-RFC complex binds to single-stranded and primed DNAs and has weak ATPase activity that is stimulated by the presence of primed DNA, replication protein A (RPA) and by proliferating cell nuclear antigen (PCNA). The CTF18-RFC complex catalyzes the ATP-dependent loading of PCNA onto primed and gapped DNA. Interacts with and stimulates DNA polymerase POLH. During DNA repair synthesis, involved in loading DNA polymerase POLE at the sites of local damage (PubMed:20227374).

CTF18 also known as Chromosome Transmission Fidelity 18 Homolog is a protein involved in DNA replication and repair. It has a molecular weight of approximately 112 kDa. CTF18 can be found in the nucleus of the cell where it interacts with other factors essential for maintaining genome integrity. Researchers identify CTF18 as a subunit of the Ctf18-RFC (Replication Factor C) complex which is key in the loading of PCNA (Proliferating Cell Nuclear Antigen) onto DNA during replication.
CTF18 plays a significant role in chromosome cohesion during cell division. It forms part of the Ctf18-RFC complex which assists in the establishment of sister chromatid cohesion. This protein complex is important for the proper segregation of chromosomes during mitosis preventing genomic instability. The coordination and function of CTF18 with other cohesin complex proteins ensure the accurate distribution of genetic material into daughter cells.
Studies show CTF18 involvement in the DNA damage response and repair pathways. It operates in tandem with proteins like RAD17 and RFC1 participating in DNA damage checkpoint activation and facilitating repair processes. CTF18 links directly to cell cycle control pathways contributing to the maintenance of genomic stability by supporting the DNA replication machinery’s response to damage and stress.
Researchers associate CTF18 with genomic instability-related conditions such as cancer and chromosomal instability syndromes. Alterations or deficiencies in CTF18 expression or function can lead to improper chromosome segregation and an increased risk of tumorigenesis. The connection between CTF18 and proteins like RAD51 which is pivotal for homologous recombination repair highlights the importance of CTF18 in maintaining cellular health.
