Biological function summary
CTLA-4 plays a critical role in downregulating immune responses. The protein forms part of a receptor complex that transmits inhibitory signals to T cells upon ligand binding. CTLA-4 inhibits T cell proliferation and cytokine production therefore preventing overactivation of the immune system. It contributes to maintaining self-tolerance and preventing autoimmunity by modulating immune checkpoints that are essential for eluding excessive immune responses.
Pathways
Several signaling events are impacted by CTLA-4 which includes its involvement in the immune checkpoint pathway and the negative regulation of T cell receptor (TCR) signaling. The interaction of CTLA-4 with B7 molecules results in the recruitment of phosphatases such as SHP-2 that dephosphorylate signaling molecules downstream of the TCR therefore attenuating T cell activation. CTLA-4 is functionally related to proteins like PD-1 another immune checkpoint protein which further modulates pathways critical for immune equilibrium.
