Immunocytochemistry/ Immunofluorescence - Anti-CTNNA1 antibody [1G5] (AB231306)
ab231306 staining CTNNA1 in A431 cells. The cells were fixed with 100% methanol (5min), permeabilized with 0.1%PBS-Tween for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab231306 at 5μg/ml and ab6046, Rabbit polyclonal to beta Tubulin - Loading Control, at 1/1000 dilution. Cells were then incubated with ab150117, Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) at 1/1000 dilution (shown in green) and ab150084, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 594) at 1/1000 dilution (shown in pseudocolor red). Nuclear DNA was labelled with DAPI (shown in blue).
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Western blot - Anti-CTNNA1 antibody [1G5] (AB231306)
This data was developed using the same antibody clone in a different buffer formulation (ab231306).
ab231306 was shown to specifically react with CTNNA1 (α-catenin) in wild type HAP1 cells. No band was observed when CTNNA1 (α-catenin) knockout samples were used. Wild-type and CTNNA1 (α-catenin) knockout samples were subjected to SDS-PAGE. ab231306 and ab181602 (Rabbit anti-GAPDH loading control) were incubated overnight at 4°C at a 1ug/ml concentraion and 1/10000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

Biological function summary
CTNNA1 is important for maintaining cellular integrity and function. It is a part of the adherens junction complex which is critical for the creation of stable cell-cell junctions in tissues. Through the adherens junction it directly associates with E-cadherin and indirectly interacts with the actin cytoskeleton. This structural configuration allows epithelial cells to remain tightly connected facilitating signal transmission between cells for coordinated tissue functioning.
Pathways
CTNNA1 plays a significant role in the Wnt signaling pathway and the regulation of actin dynamics. In the Wnt pathway CTNNA1 is associated with cell fate determination and is known to interact with beta-catenin an important component of this signaling cascade. Additionally by influencing actin cytoskeleton organization CTNNA1 affects cell movement and signal transduction essential for processes like embryogenesis and wound healing.
