Supplementary information

CUEDC2 also known as CUE domain-containing 2 is an intracellular protein with a molecular mass of approximately 45 kDa. It has a CUE domain which often associates with ubiquitin-binding. This protein is widely expressed in various tissues notably in the liver placenta and kidney. As a component of cellular machinery CUEDC2 plays a significant role in modulating cellular functions.
Biological function summary

The functions of CUEDC2 touch on several cellular processes including inflammation and cell cycle regulation. The protein can modulate signal transduction pathways by influencing transcription factors. It interacts with the anaphase-promoting complex/cyclosome (APC/C) controlling the ubiquitination and degradation of cell cycle proteins. CUEDC2 through these interactions influences the maintenance of cell cycle checkpoints and contributes to the controlled progression of cells through different stages of the cell cycle.

Pathways

CUEDC2 is integral to the NF-kappaB signaling pathway which is a critical mediator of inflammatory responses. CUEDC2 also participates in the c-Jun N-terminal kinase (JNK) pathway. In these contexts CUEDC2 interacts with proteins such as IKK and JNK to regulate the activation and suppression of downstream targets. These interactions highlight its role in maintaining cellular homeostasis and responding to external stimuli.

Researchers associate CUEDC2 with cancer and inflammatory diseases. In cancer the protein modulates pathways critical for tumor growth and invasion suggesting a possible role in tumorigenesis. CUEDC2's interactions with proteins like IKK and components of the APC/C link it to disorders characterized by aberrant cell proliferation and survival. This makes CUEDC2 a potential therapeutic target in developing treatments for related pathological conditions.

Target data

Down-regulates ESR1 protein levels through the ubiquitination-proteasome pathway, regardless of the presence of 17 beta-estradiol. Also involved in 17 beta-estradiol-induced ESR1 degradation. Controls PGR protein levels through a similar mechanism.
