Immunohistochemical analysis of paraffin-embedded (A) HEK-293T (human embryonic kidney epithelial cell) transfected with a mouse CXCL1 expression vector containing a Myc-His tag. (B) HEK-293T transfected with a mouse CXCL2 expression vector containing a Myc-His tag. (C) HEK-293T transfected with empty vector containing a Myc-His tag. tissue labeling CXCL1/GRO alpha with ab322200 at 1/2000 (0.261 ug/ml) dilution, followed by a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection).
Positive staining on HEK-293T transfected with a Myc-His-tagged mouse CXCL1 construct (image A). No staining on HEK-293T transfected with a Myc-His-tagged mouse CXCL2 construct (image B). No staining on HEK-293T transfected with empty plasmid (image C). The section was incubated with ab322200 for 30 mins at room temperature.
The immunostaining was performed on a Leica Biosystems BOND® RX instrument
Counterstained with Hematoxylin.
Secondary antibody only control : Secondary antibody is a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection).
Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins

Low expression tissue : no staining on mouse liver. The section was incubated with ab322200 for 30 mins at room temperature.
The immunostaining was performed on a Leica Biosystems BOND® RX instrument
Counterstained with Hematoxylin.

Biological function summary
CXCL1 functions as a significant mediator in inflammatory processes. It does not form a complex but acts independently to exert its effects. By attracting neutrophils to sites of injury or infection CXCL1 supports the body's defense mechanisms. This chemokine also influences angiogenesis contributing to the formation of new blood vessels which is essential in wound healing and tissue regeneration.
Pathways
CXCL1 plays a role in both the NF-kB signaling and MAPK signaling pathways. These pathways are pivotal for the regulation of immune and inflammatory responses. CXCL1 acts alongside proteins such as IL-8 another chemokine that also binds to the CXCR2 receptor. Through these pathways CXCL1 influences the activation and migration of immune cells facilitating a rapid response to inflammatory stimuli.
