In Western blot, ab238081 was shown to bind specifically to Cyclin E1. A band was observed at 47 kDa in wild-type HAP1 cell lysates with no signal observed at this size in CCNE1 CRISPR-Cas9 edited HAP1 cell line. The band observed in the CRISPR-Cas9 edited lysate lane below 47 kDa is likely to represent a truncated form of Cyclin E1. This has not been investigated further and the functional properties of the gene product have not been determined. To generate this image, wild-type and CCNE1 CRISPR-Cas9 edited HAP1 cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in fluorescent western blot (TBS-based) blocking solution before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Mouse IgG H&L (IRDye® 800CW) preabsorbed (ab216772) and Goat anti-Rabbit IgG H&L (IRDye® 680RD) preabsorbed (ab216777) at 1/20000 dilution.

Analysis of Protein Array containing more than 19,000 full-length human proteins using ab238081.Z- and S- Score : The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProtTM array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProtTM are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target. A MAb is considered to specific to its intended target, if the MAb has an S-score of at least 2.5. For example, if a MAb binds to protein X with a Z-score of 43 and to protein Y with a Z-score of 14, then the S-score for the binding of that MAb to protein X is equal to 29.
