The Cytochrome b245 Light Chain also known as p22-phox or CYBA plays a significant role in the NADPH oxidase complex. This protein has a molecular mass of approximately 22 kDa and is widely expressed in various cell types particularly in those associated with the immune system such as neutrophils. p22-phox forms part of the membrane-bound flavocytochrome b558 along with gp91-phox which is important for the generation of reactive oxygen species.

The p22-phox serves as a core component of the NADPH oxidase complex which is integral to cellular responses to infection and stress. It operates in conjunction with other cytosolic subunits to produce reactive oxygen species that help in microbial destruction and signal transduction. 4% formaldehyde-fixed, 0.2% Triton X-100 permabilized MCF7 (human breast adenocarcinoma cell line) cells stained for Cytochrome b245 Light Chain/p22-phox (green) using ab236295 at 1/66 dilution in ICC/IF, followed by Alexa Fluor 488® conjugated Goat Anti-Rabbit IgG (H+L). Counter-stained with DAPI.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
