IHC image of Cytochrome b245 Light Chain/p22-phox staining in a section of formalin-fixed paraffin-embedded normal human lung* performed on a Leica BONDTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20mins. The section was then incubated with ab75941, 1ug/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Immunohistochemical analyses of p22phox using ab75941 (1/200) in normal adrenocortical tissue (b-c) and adrenal aldosterone-producing adenoma, (e-f) p22phox presents a gradient of expression in the entire adrenal cortex (100x), and its localization was restricted to the cell membrane (400x). Sections were incubated with primary antibody at 4°C overnight and with a HRP-conjugated secondary antibody for 30 minutes at 37°C.

The Cytochrome b245 Light Chain also known as p22-phox or CYBA plays a significant role in the NADPH oxidase complex. This protein has a molecular mass of approximately 22 kDa and is widely expressed in various cell types particularly in those associated with the immune system such as neutrophils. p22-phox forms part of the membrane-bound flavocytochrome b558 along with gp91-phox which is important for the generation of reactive oxygen species.
Biological function summary

Subunit of NADPH oxidase complexes that is required for the NADPH oxidase activity that generates, in various cell types, superoxide from molecular oxygen utilizing NADPH as an electron donor (By similarity). Subunit of the phagocyte NADPH oxidase complex that mediates the transfer of electrons from cytosolic NADPH to O2 to produce the superoxide anion (O2(-)). In the activated complex, electrons are first transferred from NADPH to flavin adenine dinucleotide (FAD) and subsequently transferred via two heme molecules to molecular oxygen, producing superoxide through an outer-sphere reaction. Activation of the NADPH oxidase complex is initiated by the assembly of cytosolic subunits of the NADPH oxidase complex with the core NADPH oxidase complex to form a complex at the plasma membrane or phagosomal membrane. This activation process is initiated by phosphorylation dependent binding of the cytosolic NCF1/p47-phox subunit to the C-terminus of CYBA/p22-phox. Aassociates with NOX3 to form a functional NADPH oxidase constitutively generating superoxide (By similarity).
