Immunocytochemistry/ Immunofluorescence analysis of HaCaT (human skin keratinocyte) cells labeling Cytokeratin 8 with purified ab227644 at 1 : 25 (5.48 µg/ml). Cells were fixed in 4% paraformaldehyde and permeabilized with 0.1% Triton X-100. Cells were counterstained with ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1 : 200 (2.5 µg/ml). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1 : 1000 (2 µg/ml) dilution. DAPI nuclear counterstain. Immunohistochemical analysis of 4% PFA-fixed, 0.2% Triton X-100 permeabilized frozen mouse small intestine tissue labeling Cytokeratin with ab227644 at 1/20 (6.85 μg/ml) dilution followed by ab150077 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) at 1/1000 dilution (2 μg/ml) (Green). The immunostaining was performed on a Leica Biosystems BOND® RX instrument. The section was incubated with ab227644 for 60 mins at room temperature. Confocal image showing positive staining on mouse small intestine tissue. The nuclear counterstain was DAPI (Blue). Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
Secondary antibody only control : Primary diluent was used instead of primary antibody, followed by secondary antibody ab150077 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) at 1/1000 dilution.
