This antibody recognizes the keratin polypeptide of 45, 46, and 56.5 kD. It shows a broad pattern of reactivity with human epithelial tissues from simple glandular epithelia to stratified squamous epithelia, like epidermis, mammary gland ducts, and tracheal epithelium.
This product was changed from ascites to tissue culture supernatant on 28 Aug 2019. Lot numbers higher than GR3272704 are from tissue culture supernatant. Cytokeratins also known by the abbreviation 'CK' are a class of intermediate filament proteins providing structural support to epithelial cells. Their mass ranges between 40 kDa to 68 kDa. Cytokeratins are expressed in epithelial tissues including the outer layer of skin and line organs such as the gut and respiratory tract. Commonly referenced with specific subtypes CK MNF116 or CK cocktail antibodies are used for detection in laboratory settings. These proteins help maintain cell integrity and protect from mechanical stress.
Keratins function as a complex to create a stable cytoskeletal network inside epithelial cells. They interact with proteins to form keratin filaments contributing to the mechanical strength and resilience of cells. Cytokeratins also play a role in intracellular signaling and cellular differentiation. They participate in wound healing processes through modulation of cell proliferation and apoptosis. The CK MNF116 marker is especially noted for its application in identifying and characterizing epithelial proteins in pathology.

Overlay histogram showing A431 cells stained with ab82612 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab82612, 2μg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (Goat Anti-Mouse IgG H&L (DyLight® 488) preadsorbed ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (Mouse IgG1, Kappa Monoclonal [B11/6] - Isotype Control ab91353, 2μg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.
