Lanes 1-3 : 20 seconds Lane 4 : 3 minutes.

Lanes 1-3 used the 6% gel and lane 4 used the 4-20% gradient gel.

The expression profile/ molecular weight observed is consistent with what has been described in the literature (PMID : 31566294 )

Western blot - Anti-DAG1 antibody [IIH6C4] (AB234587)

ab234587 was shown to react with DAG1 in wild-type HAP1 cells in Western blot with loss of signal observed in a DAG1 knockout cell line. Wild-type HAP1 and DAG1 knockout cell lysates were subjected to SDS-PAGE. Membranes were blocked in 5% milk in TBST for 1 hr before incubation with ab234587 overnight at 4 °C at a 1/500 dilution. Blots were incubated with secondary antibodies at 0.2ug/mL before imaging.

The dystroglycan complex is involved in a number of processes including laminin and basement membrane assembly, sarcolemmal stability, cell survival, peripheral nerve myelination, nodal structure, cell migration, and epithelial polarization.. Alpha-dystroglycan. Extracellular peripheral glycoprotein that acts as a receptor for extracellular matrix proteins containing laminin-G domains. Receptor for laminin-2 (LAMA2) and agrin in peripheral nerve Schwann cells. Also acts as a receptor for laminin LAMA5 (By similarity).. Beta-dystroglycan. Transmembrane protein that plays important roles in connecting the extracellular matrix to the cytoskeleton. Acts as a cell adhesion receptor in both muscle and non-muscle tissues. Receptor for both DMD and UTRN and, through these interactions, scaffolds axin to the cytoskeleton. Also functions in cell adhesion-mediated signaling and implicated in cell polarity..
