Immunocytochemistry/ Immunofluorescence - Anti-DAP12 (phospho Y91) antibody [EPR25630-43] - BSA and Azide free (AB314892)

Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized 293T (human embryonic kidney epithelial cell) cells labelling DAP12 (phospho Y91) with ab314891 at 1/100 (5.04 ug/ml) dilution, followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 (2 ug/mL) dilution (Green). Confocal image showing positive staining in 293T cells transfected with a human TYROBP expression vector containing a Myc tag, while no staiing in 293T cells transfected with a human mutant TYROBP (Y91F) expression vector. All the cells are treated with pervanadate (50 uM) for 30 min. Image was taken with a confocal microscope(Leica-Microsystems, TCS SP8). Myc-Tag Mouse mAb (Alexa Fluor® 647) was used to counterstain at 1/100 (0.38ug/ ml) dilution (Red). The Nuclear counterstain was DAPI (Blue).
Secondary antibody only control : Secondary antibody is ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed at 1/1000 (2 ug/mL) dilution.

Confocal image showing cytoplasmic staining in THP-1 cells treated with pervanadate (1 mM) for 1 hr, and no staining after phosphatase treatment at 37℃ for 2 hr. Image was taken with a confocal microscope(Leica-Microsystems, TCS SP8). ab195889 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) was used to counterstain tubulin at 1/200 (2.5ug/ ml) dilution (Red). The Nuclear counterstain was DAPI (Blue).
