Dbx2 also known as developing brain homeobox 2 is a homeodomain-containing transcription factor. This protein has an approximate mass of 37 kDa. Dbx2 is mainly expressed in the developing central nervous system particularly in the neural tube where it plays a role in determining neuronal cell fate. During early development dbx2 participates in the regulation of gene expression that dictates the positioning and specialization of neurons.
Biological function summary

The transcription factor Dbx2 contributes to the differentiation of neuronal progenitor cells. Although not part of a multi-protein complex it interacts with other transcription factors to influence the transcriptional networks required for neurogenesis. Dbx2 orchestrates the precise development of specific neuron types by binding to the regulatory regions of genes responsible for neural differentiation.

Pathways

Dbx2 integrates into the pathways responsible for neural patterning and differentiation particularly the Wnt signaling pathway and the Sonic hedgehog (Shh) pathway. In these pathways Dbx2 interacts with proteins like Gli3 in the Shh pathway to modulate the transcriptional outcomes necessary for the development of the dorsoventral axis in the spinal cord. Dbx2 ensures correct levels of activity in these pathways facilitating organized neurodevelopment.

Research links Dbx2 to developmental abnormalities such as spina bifida a condition resulting from improper closure of the neural tube. Studies also suggest potential connections between alterations in Dbx2 expression and certain neurodevelopmental disorders including autism spectrum disorder. Dbx2’s interactions with Shh pathway components like Gli1 highlight its potential involvement in these disorders through misregulation of critical developmental signals.
