DDAH2 or Dimethylarginine Dimethylaminohydrolase 2 is an enzyme encoded by the human DDAH2 gene. With a molecular mass of approximately 30 kDa DDAH2 plays key role in the regulation of nitric oxide (NO) levels by breaking down asymmetric dimethylarginine (ADMA) an endogenous inhibitor of nitric oxide synthase (NOS). DDAH2 expression is found in a variety of tissues including the heart liver and kidney indicating its widespread functional importance in these organs.
Biological function summary

DDAH2 regulates nitric oxide production affecting processes such as vasodilation and vascular homeostasis. This enzyme also interacts in complex regulatory networks that modulate blood flow and blood pressure. By controlling ADMA concentration DDAH2 indirectly influences endothelial function and cardiovascular health. The enzyme’s role in maintaining NO levels makes it integral to processes that rely on proper endothelial signaling.

Pathways

Nitric oxide signaling and the methylarginine metabolic pathways are two significant areas involving DDAH2. In the NO signaling pathway DDAH2 impacts the synthesis of NO by modulating inhibitor levels of NOS. In the methylarginine pathway DDAH2 is closely related to the enzyme PRMT (Protein Arginine Methyltransferase) as it regulates the availability of methylarginine substrates. DDAH2’s activity harmonizes the balances within these pathways ensuring effective signaling and metabolic regulation.

Altered DDAH2 activity has connections to cardiovascular diseases and hypertension. Dysfunctional DDAH2 can lead to elevated ADMA levels which correlate with increased cardiovascular risk and impaired endothelial function. Within these diseases DDAH2’s interaction with NOS is important; improper NO production resulting from dysregulated enzyme activity affects cardiovascular health. Addressing DDAH2 malfunction holds therapeutic potential for managing related vascular disorders.

Putative hydrolase with unknown substrate (Probable). Does not hydrolyze N(G),N(G)-dimethyl-L-arginine (ADMA) which acts as an inhibitor of NOS (PubMed : 21493890, PubMed : 37296100). In endothelial cells, induces expression of vascular endothelial growth factor (VEGF) via phosphorylation of the transcription factor SP1 by PKA in a process that is independent of NO and NO synthase (By similarity). Similarly, enhances pancreatic insulin secretion through SP1-mediated transcriptional up-regulation of secretagogin/SCGN, an insulin vesicle docking protein (By similarity). Upon viral infection, relocates to mitochondria where it promotes mitochondrial fission through activation of DNM1L leading to the inhibition of innate response activation mediated by MAVS (PubMed : 33850055).
