The DDHD1 protein also known as DDH1 or PA-PLA1 is a phospholipase involved in lipid metabolism. With a molecular mass of approximately 88 kDa this protein localizes primarily in the cytosol but can also associate with the Golgi apparatus. DDHD1 is expressed in various tissues with significant levels in the brain and testis. This expression pattern suggests its potential role in central nervous system functions and reproductive processes.
Biological function summary

DDHD1 enzyme is responsible for the hydrolysis of phosphatidic acid contributing to lysophospholipid production. It is part of the intracellular membrane trafficking system operating independently rather than as part of a complex. By modulating the lipid composition of membranes DDHD1 helps regulate membrane curvature and dynamics which affects cellular processes such as vesicular transport and signaling.

Pathways

DDHD1 plays a significant role in the phospholipid signaling pathway by acting on phosphatidic acids. Its activity influences the levels of signaling molecules like lysophosphatidic acid which impact cellular events like proliferation and migration. Additionally DDHD1 is involved in the arachidonic acid pathway connecting it with enzymes such as PLA2 which also mediate the release of fatty acids from phospholipids essential for inflammatory responses.

DDHD1 mutations have been associated with hereditary spastic paraplegia a neurodegenerative condition characterized by progressive weakness and spasticity of the lower limbs. This relationship indicates its contribution to maintaining neuronal integrity. Furthermore DDHD1 function has links to mitochondrial lipid homeostasis associating it with DDHD2 in the context of complex lipid synthesis disorders especially those affecting the nervous system.

Phospholipase A1 (PLA1) that hydrolyzes ester bonds at the sn-1 position of glycerophospholipids producing a free fatty acid and a lysophospholipid (Probable) (PubMed : 20359546, PubMed : 22922100).
