DDX18 also known as DEAD-box helicase 18 functions as an RNA helicase involved in the modification of RNA structures. It unwinds RNA and is important in ribosome biogenesis. The molecular weight of DDX18 is approximately 73 kDa. DDX18 is expressed in various tissues with significant presence in the nucleolus of cells where ribosome production occurs.
Biological function summary

DDX18 plays a role in the assembly and maturation of pre-ribosomal particles. It functions within a multi-protein complex and interacts with other nucleolar proteins. This interaction is necessary for the conversion of pre-rRNA into mature rRNA which forms the structural and functional components of ribosomes essential for protein synthesis.

Pathways

DDX18 participates in the ribosome biogenesis pathway which is key to cellular protein production. It is involved in the nucleolar preribosome process alongside proteins like NOP58 and fibrillarin. DDX18 contributes to the processing of pre-rRNA intermediates critical for creating functional ribosomes which are necessary for cell growth and proliferation.

Mutations or dysregulation of DDX18 associate with certain cancers like colorectal cancer. These changes impact ribosomal RNA processing and consequently protein synthesis leading to uncontrolled cell growth. DDX18's interaction with c-Myc a protein involved in cell cycle regulation and growth highlights its connection to cancer pathology.
