The DDX23 protein also known as DEAD-box helicase 23 is a member of the DEAD-box protein family. This family is characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD) and is involved in a wide range of cellular processes. DDX23 plays a mechanical role in unwinding RNA which is essential for RNA processing. The protein has a molecular mass of approximately 82 kDa. DDX23 is expressed ubiquitously across various tissues with significant expression in the nucleus where it contributes to RNA dynamics.
Biological function summary

DEAD-box helicase 23 participates in the spliceosome complex important for pre-mRNA splicing a process that removes introns and joins exons to form mature messenger RNA. DDX23 acts during spliceosome assembly and the catalytic activation phase. Through its helicase activity it facilitates structural transitions within the spliceosome ensuring accurate splicing of pre-mRNA molecules. This function is essential for accurate gene expression and cellular function.

Pathways

DDX23 is involved in the spliceosomal pathway and related RNA processing pathways. Its role in pre-mRNA splicing links DDX23 to the regulation of gene expression impacting numerous downstream pathways. Within these pathways DDX23 interacts closely with other spliceosome-associated proteins like PRPF8 and SNRNP200. These interactions ensure the efficiency and regulation of the splicing process critical to cellular and physiological functions.

Alterations in the DDX23 function have been associated with conditions such as cancers and neurodegenerative diseases. Independently of its spliceosome formation function, required for the suppression of incorrect R-loops formed during transcription; R-loops are composed of a DNA : RNA hybrid and the associated non-template single-stranded DNA (PubMed : 28076779).
