DDX27 also known as DEAD-box protein 27 operates as an important RNA helicase involved in the unwinding of RNA structures. This protein with a molecular weight of approximately 85 kDa belongs to the DEAD-box family characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD). It is ubiquitously expressed in various tissues prominently in the nucleolus where it functions in ribosomal RNA (rRNA) biogenesis and assembly.
Biological function summary

DEAD-box protein 27 participates in ribosome biogenesis and interacts with the pre-rRNA processing complexes. This protein acts as an ATP-dependent helicase contributing to the modification and processing of rRNA during ribosome assembly. It often associates with several nucleolar proteins forming a part of multi-protein complexes essential for the synthesis and maturation of ribosomal subunits.

Pathways

The role of DDX27 integrates mainly within the ribosome biogenesis and RNA processing pathways. It interacts with other DEAD-box proteins such as DDX18 and DDX56 which collaborate in modulating ribosomal RNA processing and ribosome assembly. Dysregulation of DDX27 is linked to certain cancers including colorectal and hepatocellular carcinoma. Additionally involvement with proteins like MYC has been observed as MYC-driven regulation of ribosomal biogenesis highlights a potential connection between DDX27 activity and oncogenic pathways.
