The DDX41 protein also known as DEAD Box Protein 41 is a member of the DEAD-box RNA helicase family. This protein weighs approximately 82 kDa. It is involved in various RNA processing activities including alteration of RNA secondary structure during spliceosome assembly ribosome biogenesis and RNA transport. DDX41 is expressed in a wide range of tissues with particularly high levels in bone marrow and immune cells. By functioning as an RNA helicase it plays a central role in the regulation of fundamental biological processes like RNA stability and translation.
Biological function summary

DDX41 plays a significant role in the innate immune response. It acts as a cytosolic DNA sensor that is involved in the early detection of viral and bacterial DNA. DDX41 interacts with the stimulator of interferon genes (STING) adaptor protein forming an important part of the DNA sensing complex that activates type I interferon responses. By recognizing and binding to foreign DNA within the cytoplasm it helps initiate an antiviral state promoting the activation of immune responses to foreign pathogens.

Pathways

The involvement of DDX41 extends to the innate immune signaling pathway and the type I interferon production pathway. Within these pathways DDX41 interacts directly with other important proteins like STING and TBK1 contributing to the signal transduction that leads to interferon production and subsequent immune activation. These pathways are important for mounting an effective defense against infections connecting DDX41 with the broader regulatory network of proteins involved in pathogen recognition and immune response modulation.

Mutations in the DDX41 gene correlate with certain hematologic conditions including myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML). Disruptions in the normal function of DDX41 can affect its role in immune signaling potentially leading to aberrant cell growth and impaired immune functions. Additionally interactions with proteins such as STING during immune responses imply that dysregulation in DDX41 activity could elucidate further insights into other immune-related disorders.
