DDX47 was immunoprecipitated from HeLa (human epithelial cell line from cervix adenocarcinoma) whole cell lysate (1 mg for IP, 20% of IP loaded) with ab225870 at 6 μg/mg lysate. Western blot was performed from the immunoprecipitate using ab225870 at 0.4 μg/ml.

DDX47 also known as DEAD box protein 47 functions as a putative RNA helicase involved in the unwinding of RNA secondary structures. This protein weighing approximately 52 kDa contains the conserved D-E-A-D amino acid motif characteristic of DEAD box proteins. It expresses broadly across various tissues but shows significant levels in the nucleolus where it plays roles in nucleolar activities.
Biological function summary

This protein participates in ribosome biogenesis by assisting in the maturation of ribosomal RNA. DDX47 as part of ribonucleoprotein complexes facilitates the cleavage of precursor rRNA which is necessary for the production of functional ribosomal subunits. This process is essential for maintaining high rates of protein synthesis in rapidly growing cells thereby influencing cellular growth and proliferation.

Pathways

RNA metabolism and ribosome biogenesis include DDX47 as an important component. In these pathways it interacts with proteins such as NOP56 and NOLC1 which are integral to ribosomal RNA processing. Through its role in these pathways DDX47 influences transcriptional and translational regulation which are fundamental processes in cell physiology.

Alterations in DDX47 expression link to cancer and ribosomopathies. Abnormal activity or dysregulation of DDX47 can affect cell proliferation contributing to tumorigenesis.
