ab126730 staining DDX5 in the human cell line HeLa (Human cervix adenocarcinoma epithelial cell) by intracellular flow cytometry. Cells were fixed with 4% paraformaldehyde, permeabilised with 90% methanol and the sample was incubated with the primary antibody at a dilution of 1/200. A goat anti rabbit IgG (Alexa Fluor® 488) at a dilution of 1/2000 was used as the secondary antibody.

Immunocytochemistry/Immunofluorescence analysis of SH-SY5Y (human neuroblastoma) cells labelling DDX5 (green) with purified ab126730 at 1/250. Cells were fixed with 4% Paraformaldehyde and permeabilized with 0.1% Triton X-100. ab150077, Alexa Fluor® 488-conjugated goat anti-rabbit IgG (1/1000) was used as the secondary antibody. DAPI (blue) was used as a nuclear counterstain.

Secondary Only Control : PBS was used instead of the primary antibody as the negative control.

DDX5 also known as DEAD-box helicase 5 or p68 is an RNA helicase important to various RNA processing tasks. With a molecular mass of approximately 68 kDa DDX5 unwinds RNA secondary structures using energy from ATP hydrolysis. It expresses widely in human tissues with higher levels observed in regions of active cellular proliferation. The expression pattern reflects its significant involvement in pathways that require dynamic RNA structure modifications.
Biological function summary

The function of DDX5 facilitates RNA splicing transcription and ribosome biogenesis. DDX5 is a component of various RNA processing complexes interacting with other proteins to modify pre-mRNA and other RNA molecules. It plays a role in modulating gene expression and ensuring the proper processing of precursor RNA transcripts. This makes DDX5 important in cellular differentiation and development influencing stem cell renewal and pluripotency.

Pathways

DDX5 participates in the Wnt/β-catenin and p53 signaling pathways. In the Wnt/β-catenin pathway DDX5 interacts with proteins such as β-catenin to influence gene transcription involved in cell growth and differentiation. Within the p53 pathway DDX5 modulates the activity of p53 a protein known for its role in DNA damage response and apoptosis. These interactions position DDX5 at the intersection of multiple regulatory networks that govern cellular proliferation and survival.

Aberrant DDX5 expression links to cancer and neurodegenerative diseases. Overexpression or dysregulation of DDX5 has been observed in various cancers including breast cancer where it can interact with proteins such as estrogen receptor alpha promoting tumor progression. In neurodegenerative disorders particularly amyotrophic lateral sclerosis (ALS) DDX5 forms toxic aggregates with TDP-43 a protein important in RNA binding and processing. Understanding DDX5's involvement in these diseases offers insight into potential therapeutic targets and diagnostic markers.

Involved in the alternative regulation of pre-mRNA splicing; its RNA helicase activity is necessary for increasing tau exon 10 inclusion and occurs in a RBM4-dependent manner. Binds to the tau pre-mRNA in the stem-loop region downstream of exon 10. The rate of ATP hydrolysis is highly stimulated by single-stranded RNA. Involved in transcriptional regulation; the function is independent of the RNA helicase activity. Transcriptional coactivator for androgen receptor AR but probably not ESR1. Synergizes with DDX17 and SRA1 RNA to activate MYOD1 transcriptional activity and involved in skeletal muscle differentiation. Transcriptional coactivator for p53/TP53 and involved in p53/TP53 transcriptional response to DNA damage and p53/TP53-dependent apoptosis. Transcriptional coactivator for RUNX2 and involved in regulation of osteoblast differentiation. Acts as a transcriptional repressor in a promoter-specific manner; the function probably involves association with histone deacetylases, such as HDAC1.
