Supplementary information

DDX53 also known as CT26 is a member of the DEAD box protein family characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD). This protein functions primarily as an ATP-dependent RNA helicase facilitating the unwinding of RNA structures. The molecular weight of DDX53 is approximately 58 kDa. Its expression is found mainly in various tumor tissues making it a cancer/testis antigen. It is not commonly expressed in normal tissues except for the testis highlighting its potential as a biomarker for cancer diagnostics.
Biological function summary

DDX53/CT26 plays a role in cellular processes related to cell growth and proliferation. It participates in mRNA metabolism and splicing though it is not necessarily part of a known stable complex. The role of DDX53 in mRNA processing links it to the regulation of gene expression at a post-transcriptional level influencing cell cycle control and proliferation dynamics. Such regulatory functions are critical in understanding how aberrant expression might contribute to tumorigenesis.

Pathways

DDX53 is involved in pathways that regulate gene expression and proliferation such as the mRNA splicing pathway. Its interaction with other RNA-binding proteins might modulate splicing events important for protein expression patterns that sustain cell growth. DDX53 relates to proteins involved in the regulation of RNA metabolism such as other DEAD box family proteins which ensure accurate splicing and mRNA turnover. By affecting these pathways DDX53 can impact cellular growth control mechanisms.

DDX53's aberrant expression is linked to certain cancers most prominently in lung cancer and melanoma. Its expression correlates with tumor presence and progression. The interaction of DDX53 with testis-specific proteins in tumors makes it a relevant target for cancer immunotherapy. Furthermore its specificity to cancerous tissues over normal ones barring the testis positions DDX53 as a potentially valuable target for cancer vaccine development leveraging immune responses against tumor cells expressing this antigen.
