Immunofluorescent analysis of 4% paraformaldehyde-fixed 0.1% Triton X-100 permeabilized Neuro-2a (Mouse neuroblastoma cells) cells labeling delta-catenin with ab184917 at 1/500 dilution followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody at 1/400 dilution (green). Cytoplasmic and nuclear staining on Neuro-2a cell line is observed. The nuclear counter stain is DAPI (blue). Tubulin is detected with ab7291 (anti-Tubulin mouse mAb) at 1/500 dilution and ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution (red).
The negative controls are as follows :
-ve control 1 : ab184917 at 1/500 dilution followed by ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution.
-ve control 2 : ab7291 (anti-Tubulin mouse mAb) at 1/500 dilution followed by ab150077 (Alexa Fluor®488 Goat Anti-Rabbit IgG H&L) at 1/400 dilution.

Delta-catenin also known as CTNND2 or Neurojungin is an important protein involved in cellular adhesion processes. With a molecular mass of around 122 kDa Delta-catenin is found mainly in the brain and nervous system tissues. It is a member of the armadillo/β-catenin superfamily and plays a significant role in the structure and function of synapses connecting cytoskeletal components to cell junctions. Its expression is most prominent in neurons suggesting a function important to neuronal development and maintenance.
Biological function summary

Delta-catenin associates with and is part of the cadherin-catenin complex facilitating cell-cell adhesion in neuronal cells. This complex supports the stabilization of adherens junctions influencing cell shape signaling and gene transcription. Delta-catenin's interaction with cadherin proteins helps regulate synaptic plasticity and dendritic spine development impacting learning and memory processes within the brain.

Pathways

Delta-catenin contributes significantly to the Wnt signaling pathway. It plays a role in cellular edge interactions and migration by modulating cadherin-mediated adhesion and Rho GTPase signaling. In collaboration with proteins such as β-catenin Delta-catenin influences processes linked to cell morphological changes and neurodevelopmental regulation. It acts as a mediator between intracellular signaling and intercellular communication impacting neurogenesis.

Delta-catenin has potential links to neurodevelopmental disorders like autism spectrum disorder (ASD) and Alzheimer's disease. Alterations in Delta-catenin expression and mutations correlate with cognitive dysfunction and synaptic impairments observed in these conditions. In Alzheimer's disease Delta-catenin's interaction with β-amyloid and presenilin-1 proteins can exacerbate synaptic loss and memory deficits identifying it as a possible target in understanding disease mechanisms and therapeutic intervention.

Involved in the regulation of canonical Wnt signaling (By similarity). It probably acts on beta-catenin turnover, facilitating beta-catenin interaction with GSK3B, phosphorylation, ubiquitination and degradation (PubMed : 20623542).
