Supplementary information

Dengue virus NS2B also known as non-structural protein 2B forms a part of the dengue virus polyprotein. It functions as a cofactor for the NS3 protease helping in the proteolytic processing of the viral polyprotein. The protein has a mass of around 14 kDa. NS2B localizes to the endoplasmic reticulum of infected host cells where it helps in transitioning viral proteins to their mature forms.
Biological function summary

NS2B collaborates with NS3 to form the NS2B-NS3 complex essential for viral replication and assembly. The NS2B-NS3 protease complex cleaves the polyprotein into individual functional viral proteins necessary for dengue virus replication. This process facilitates the viral lifecycle within the host cells by allowing successive steps in viral replication processes.

Pathways

NS2B coordinates its activities with the NS3 protein within the viral replication and assembly pathway. This pathway involves viral genome RNA replication and assembly of the virions. NS2B-NS3 complex activity results in efficient replication of the dengue virus RNA facilitating the production of virus particles. In this process NS2B may interact with other viral proteins like NS5 which plays a role in RNA synthesis and capping serving as an interconnected pathway for successful viral propagation.

NS2B is directly linked to the pathogenesis of dengue fever and dengue hemorrhagic fever. The activity of the NS2B-NS3 protease complex is important for the viral life cycle making it an important target for antiviral drug development. Dysregulation of NS2B function leads to uncontrolled viral replication which is a primary factor in dengue virus infection. The complex relationship of NS2B with other viral proteins highlights its potential as a focal point in therapeutic intervention strategies against dengue virus-induced diseases.
