Non-infected and infected BHK-21 whole cell extracts (5 ug) were separated by 7.5% SDS-PAGE and the membrane was blotted with ab317175 diluted at 1 : 200000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

Dengue Virus NS3 also known as NS3 protein is a non-structural protein with enzyme activity mainly acting as a serine protease and RNA helicase. Weighing approximately 69 kDa NS3 is expressed in infected cells of vertebrate hosts and is an important component in the virus replication process. It requires NS2B as a cofactor to display its protease activity facilitating the cleavage of the viral polyprotein precursor into functional proteins.
Biological function summary

NS3 protein plays a significant role in viral replication and maturation by partaking in the intracellular processes of the dengue virus. NS3 acts in concert with other non-structural proteins to ensure efficient replication of the viral genome. Particularly the helicase function assists in unwinding RNA during genome synthesis making it indispensable for the viral life cycle.

Pathways

NS3 protein participates in the dengue virus replication and maturation pathways which are critical for viral propagation. It interacts directly with other dengue virus proteins like NS2B and NS5 the latter functioning as the viral polymerase. NS3 and NS5 establish a multi-protein replicase complex that facilitates RNA genome replication and processing.
