We have systematically measured KD (the equilibrium dissociation constant between the antibody and its antigen), of more than 840 recombinant antibodies to assess not only their individual KD values but also to see the average affinity of antibody. Based on the comparison with published literature values for mouse monoclonal antibodies, Recombinant antibodies appear to be on average 1-2 order of magnitude higher affinity.

Product details

ab248688 is the carrier-free version of ab134117.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

The DENN domain-containing proteins also referred to as DENN serve as specific guanine nucleotide exchange factors (GEFs) that activate Rab GTPases important for vesicular trafficking. These proteins generally weigh between 110 to 150 kDa. DENN proteins predominantly are found within cellular membranes including endosomes. This positioning is consistent with their involvement in vesicle trafficking and membrane dynamics.
Biological function summary

DENN domain proteins facilitate critical processes such as endocytic recycling autophagy and transport processes within cells. These proteins belong to a complex containing multiple subunits that coordinate the activation of Rab GTPases. Through these activities DENN proteins ensure proper membrane trafficking dynamics and cellular organization integral to maintaining cell function.

Pathways

DENN proteins participate in cellular pathways like endocytic recycling and autophagy. In the endocytic pathway DENN proteins regulate Rab GTPases such as Rab5 and Rab7 controlling vesicle formation and movement. They also play a part in mTOR signaling influencing cell growth and autophagy regulation. By interacting with these key pathways DENN domain proteins help to integrate signals and coordinate cellular responses to environmental changes.

Alterations in DENN proteins often relate to neurodegenerative disorders and cancer. Mutations in the DENN domain have links to conditions such as Parkinson’s disease where Rab GTPases affected are implicated in neuronal function and survival. In cancer dysregulation of DENN protein function can influence cellular proliferation and apoptosis affecting tumor development. DENN proteins through their interactions and related pathways represent significant factors in understanding and potentially addressing these conditions.

Target data

Guanyl-nucleotide exchange factor that regulates small GTPases of the Rab family (PubMed : 18559336, PubMed : 20937701). Converts GDP-bound inactive form of RAB27A and RAB27B to the GTP-bound active forms (PubMed : 18559336, PubMed : 20937701). Converts GDP-bound inactive form of RAB3A, RAB3C and RAB3D to the GTP-bound active forms, GTPases involved in synaptic vesicle exocytosis and vesicle secretion (By similarity). Plays a role in synaptic vesicle formation and in vesicle trafficking at the neuromuscular junction (By similarity). Involved in up-regulating a post-docking step of synaptic exocytosis in central synapses (By similarity). Probably by binding to the motor proteins KIF1B and KIF1A, mediates motor-dependent transport of GTP-RAB3A-positive vesicles to the presynaptic nerve terminals (By similarity). Plays a role in TNFA-mediated activation of the MAPK pathway, including ERK1/2 (PubMed : 32761064). May link TNFRSF1A with MAP kinase activation (PubMed : 9115275). May be involved in the regulation of TNFA-induced apoptosis (PubMed : 11577081, PubMed : 32761064).
