Desmoplakin is a critical component of the cell structure acting as a link between desmosomes and the intermediate filament network. It exists in two isoforms Desmoplakin I and Desmoplakin II with molecular masses of approximately 332 kDa and 260 kDa respectively. Desmoplakin is mainly expressed in tissues with high mechanical stress such as the heart and skin where strong cell adhesion is necessary. Alternate names like Desmoplakine may also be used in some scientific literature.
Desmoplakin is essential for maintaining cell adhesion and structural integrity within tissues. It serves as part of the desmosomal complex where it anchors intermediate filaments to the desmosomal plaque ensuring the mechanical stability required by tissues under mechanical stress. This function is particularly critical in epithelial cells and cardiac tissue where desmoplakin interacts closely with other desmosomal components to maintain robust cell-to-cell connections.
Desmoplakin plays a significant role in the regulation of adhesive structures through its involvement in the intercellular junctional complex pathways. The protein collaborates with other key components of these pathways such as plakoglobin and plakophilins. Its role in these pathways ensures that cell adhesion and stability are maintained across different tissue types which is vital for normal cellular function and organization.
Mutations or dysfunctional interactions of desmoplakin can lead to conditions like arrhythmogenic right ventricular cardiomyopathy (ARVC) and skin disorders such as epidermolysis bullosa simplex. In ARVC desmoplakin-related defects may disrupt interactions with other desmosomal proteins like plakoglobin impacting heart muscle integrity and function. Similarly in epidermolysis bullosa simplex altered desmoplakin function compromises skin cell adhesion leading to increased susceptibility to injury.
