Function

Type-VI intermediate filament (IF) which plays an important cytoskeletal role within the muscle cell cytoskeleton. It forms heteromeric IFs with desmin and/or vimentin, and via its interaction with cytoskeletal proteins alpha-dystrobrevin, dystrophin, talin-1, utrophin and vinculin, is able to link these heteromeric IFs to adherens-type junctions, such as to the costameres, neuromuscular junctions, and myotendinous junctions within striated muscle cells.

Activity summary

Desmuslin also known as SYN or Synemin is an intermediate filament protein with a mass of approximately 180 kDa. It is prominently expressed in muscle tissues including skeletal cardiac and smooth muscles. Desmuslin integrates into the cytoskeleton providing essential structural support and maintaining cellular integrity. Its expression is not restricted to muscle tissues; desmuslin also appears in certain non-muscle cells indicating its diverse functional roles.

Biological function summary

The protein desmuslin plays an important role in maintaining cellular architecture and mechanical stability. It associates closely with the cytoskeletal network and participates in the actin-binding protein complex. Desmuslin interacts with various cytoskeletal components anchoring them to the sarcolemma and linking the extracellular matrix to the cytoskeleton. This interaction supports cellular resilience against mechanical stress and contributes to tissue homeostasis.

Pathways

The protein desmuslin integrates into cell adhesion and signaling pathways. It links mechanically to pathways involving integrins which facilitate cell adhesion and communication with the extracellular matrix. Through these pathways desmuslin associates with proteins like plectin and dystrophin which also contribute to structural integrity. This interaction aids in the transmission of mechanical signals integrating external stimuli with intracellular responses and supports proper muscle function.

Associated diseases and disorders

The protein desmuslin associates with conditions such as muscular dystrophy and cardiomyopathies. Mutations or dysregulation of desmuslin can disrupt muscle integrity leading to progressive muscle weakness and degeneration. Desmuslin interacts with the dystrophin-glycoprotein complex which when altered can also contribute to the development of muscular dystrophy. Additionally desmuslin plays a role in maintaining cardiac muscle function and its dysfunction relates to certain cardiomyopathies posing significant implications for cardiac health.
