Immunohistochemical analysis of formalin fixed paraffin embedded human kidney tissue labelling DHHC-12 with ab237688 at a 1 : 50 dilution. The immunostaining was performed on a Leica Bond™ system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 min at RT. ab237688 was incubated at 4°C overnight. The primary was detected using a Goat anti-rabbit polymer IgG HRP and visualized using 0.05% DAB.

The DHHC-12 protein also known as zDHHC12 functions as a palmitoyl acyltransferase enzyme facilitating the addition of palmitate to target proteins. This post-translational modification of proteins affects their trafficking stability or function. DHHC-12 has a molecular mass of approximately 39 kDa. Expression of DHHC-12 occurs in various tissues with significant levels detectable in brain and some other organs. Its activity frequently involves dynamic regulation of protein interactions and membrane associations.
Biological function summary

DHHC-12 is key in ensuring regulation of protein localization and signaling pathways by mediating protein palmitoylation. As part of a large family of palmitoyl acyltransferases it functions based on substrate specificity displaying selective enzymatic activity towards a subset of protein targets. This selectivity allows DHHC-12 to influence diverse cellular processes including signal transduction and protein trafficking. While not directly part of a multiprotein complex DHHC-12 interacts with various proteins during its palmitoylation activities.

Pathways

DHHC-12 plays important roles in cellular signaling and membrane protein trafficking. It significantly contributes to the Ras signaling pathway which controls cell proliferation differentiation and survival. Through this pathway DHHC-12 interacts with Ras proteins modulating their membrane association and activity. DHHC-12 also influences the Wnt signaling pathway which affects cell fate and tissue homeostasis. It ensures proper membrane localization of Wnt pathway components maintaining the signaling cascade integrity.

DHHC-12 has implications in neurodegenerative conditions with particular relevance to Alzheimer's disease. Altered palmitoylation patterns mediated by DHHC-12 can affect amyloid precursor protein processing or tau protein function linking it to disease pathogenesis. Additionally aberrant DHHC-12 expression or activity correlates with certain types of cancers where it affects important oncogenic pathways. Through these disease associations DHHC-12 connects to proteins such as APP in Alzheimer's disease and oncogenes in cancer highlighting its potential as a therapeutic target.
