Palmitoyltransferase that catalyzes the addition of palmitate onto various protein substrates, such as CGAS, HRAS and LCK (PubMed:23034182, PubMed:27481942, PubMed:35438208). Acts as a negative regulator of the cGAS-STING pathway be mediating palmitoylation and inactivation of CGAS (PubMed:35438208). May also have a palmitoyltransferase activity toward the beta-2 adrenergic receptor/ADRB2 and therefore regulate G protein-coupled receptor signaling (PubMed:27481942).

DHHC-18 also known as ZDHHC18 functions as a palmitoyltransferase enzyme responsible for the addition of palmitate a type of lipid to target proteins. This process is known as palmitoylation and it influences the proteins' membrane association and function. DHHC-18 has a molecular weight of about 41 kDa. Expression of DHHC-18 occurs in various tissues including the brain and heart implicating its role in diverse cellular processes.
DHHC-18 contributes to protein trafficking and signaling by influencing the membrane dynamics. It does not act alone but as part of a large dynamic network of proteins regulating lipid modifications. This network helps in processes such as synaptic signaling and neuronal growth highlighting its importance in neurobiology. Through its catalytic activity DHHC-18 can modify substrates like Ras-related proteins affecting their functional states and interactions.
DHHC-18 interacts within the Ras signaling and synaptic transmission pathways. Through its involvement in Ras signaling DHHC-18 modulates the activity of Ras proteins which contributes to cell growth and differentiation. Furthermore the synaptic transmission pathway gets influenced by this palmitoylation enzyme affecting neurotransmitter release with interacting proteins like PSD-95 playing key roles in maintaining synaptic plasticity and stability.
DHHC-18 has implications in neurological conditions such as schizophrenia and autism spectrum disorders. Its direct connection involves the modulation of neuronal membrane proteins important for proper brain function. Abnormal palmitoylation due to DHHC-18 dysfunction can disrupt the balance of synaptic signaling leading to cognitive and behavioral anomalies. Dysregulation can also influence proteins like NLGN3 and CNTNAP2 which are pivotal in psychiatric and neurodevelopmental disorders.
