Supplementary information

DHRS9 also known as dehydrogenase/reductase SDR family member 9 is an enzyme encoded by the DHRS9 gene. It belongs to the short-chain dehydrogenases/reductases (SDR) family and has a molecular mass of about 34 kDa. This protein acts mainly as an oxidoreductase involved in the metabolism of various substrates including retinoids and xenobiotics. DHRS9 has broad tissue distribution but high expression levels are noted in the liver kidney and placenta.
Biological function summary

As a member of the SDR family DHRS9 plays roles in redox reactions that are important for metabolic functions. It does not typically form complexes with other proteins but cooperates with enzymes such as retinol dehydrogenases. This protein is involved in the conversion of retinol to retinal which is a part of vitamin A metabolism that is essential for vision and cellular differentiation.

Pathways

Several significant metabolic processes involve DHRS9. It is primarily engaged in retinoic acid metabolism influencing retinoid signaling pathways which are vital for embryonic development and homeostasis. In these pathways DHRS9 interacts closely with retinoic acid receptors (RAR) and cellular retinoic acid-binding proteins which regulate gene expression required for cell differentiation and proliferation.

Research connects DHRS9 to specific conditions. Aberrant expression of DHRS9 has been associated with certain liver diseases and cancer types. In cases of liver disease misregulation of DHRS9 affects retinol metabolism impacting liver function and regenerative processes. Studies also suggest linkage between DHRS9 and RARs in oncogenic pathways where altered retinoic acid metabolism may affect tumor suppression mechanisms.

Target data

3-alpha-hydroxysteroid dehydrogenase that converts 3-alpha-tetrahydroprogesterone (allopregnanolone) to dihydroxyprogesterone and 3-alpha-androstanediol to dihydroxyprogesterone (PubMed : 11294878, PubMed : 29541409). Also plays a role in the biosynthesis of retinoic acid from retinaldehyde (PubMed : 11304534, PubMed : 12618084). Can utilize both NADH and NADPH.
