Supplementary information

DHX30 also known as DExH-Box Helicase 30 is a member of the DEAD-box helicase family and plays a role in RNA metabolism. This protein has a mass of approximately 132 kDa and is widely expressed across various tissues with notable abundance in the brain and testes. Its core mechanical function includes unwinding RNA structures which is critical for the assembly and remodeling of ribonucleoprotein complexes. By doing so DHX30 facilitates various steps in RNA processing including transcription splicing and translation initiation.
Biological function summary

Within cellular processes DHX30 contributes to mitochondrial function and protein synthesis by participating in RNA splicing and translation. It forms part of larger ribonucleoprotein complexes that regulate RNA dynamics. The protein interacts with other cellular components to coordinate the expression of genes involved in cellular growth and metabolism. Its presence in the mitochondrial matrix indicates a specific role in maintaining mitochondrial health and efficient energy production.

Pathways

The DHX30 protein influences cellular activities related to RNA-binding and translation regulation. It is deeply integrated within the translation initiation pathway working alongside RNA-binding proteins to regulate gene expression efficiently. Within this context DHX30 associates with proteins such as EIF4A which is involved in initiating translation of mRNA. Moreover DHX30 participates in maintaining proper functioning of the mitochondrial translation machinery ensuring the synthesis of essential mitochondrial proteins that power the cell's energy requirements.

DHX30 has links to intellectual disabilities and neurodegenerative disorders. Variants or deficiencies in DHX30's function can contribute to conditions such as neurodevelopmental disorders which could potentially connect with other proteins like FMRP involved in synaptic function and mental development. Additionally disruptions in DHX30 activity have been observed in certain types of cancers where aberrant RNA processing and gene expression stability are important factors. Understanding these associations may provide insights into the role of DHX30 in maintaining cellular integrity and its potential as a therapeutic target.
