Supplementary information

DHX32 also known as DExH-box helicase 32 is a member of the DEAH-box family of RNA helicases. With a mass of approximately 84 kDa DHX32 plays important roles in unwinding RNA secondary structures facilitating processes like RNA splicing and ribosome biogenesis. This protein is highly expressed in various tissues with notable abundance in lymphoid tissues suggesting its involvement in immune function. Its ATP-dependent RNA helicase activity makes it central to post-transcriptional regulation and RNA metabolism.
Biological function summary

DHX32 contributes to the regulation of RNA-related processes. By being part of larger ribonucleoprotein complexes it influences the maturation and processing of ribosomal RNA pre-mRNA splicing and other RNA metabolic pathways. Its function in these complexes ensures the proper assembly and function of the ribosomes which is essential for protein synthesis. The helicase activity provides the energy necessary for these molecular events directly impacting the efficiency and fidelity of RNA processing.

Pathways

Research has indicated that DHX32 is involved in both the spliceosome assembly pathway and the ribosome biogenesis pathway. Within these pathways DHX32 interacts with proteins such as SF3A1 and U2AF2 in the splicing machinery and nucleophosmin (NPM1) in ribosome assembly. These interactions connect DHX32 to a broader network of proteins and ensure the pathways proceed correctly affecting cellular growth and maintenance processes.

DHX32 has shown associations with lymphomas and leukemias reflecting its expression in lymphoid tissues. Its interaction with proteins like c-Myc a well-known oncogene links DHX32 to the regulation of cellular proliferation and survival. These interactions suggest that dysregulation of DHX32 can contribute to the development of malignancies by altering RNA processing and protein synthesis highlighting its potential as a target for therapeutic approaches.
