DIAPH2 also known as DIA or Diaphanous-related formin-2 is a protein that functions as a formin homology protein involved in actin polymerization. It plays an important role in the dynamic reorganization of the cytoskeleton. DIAPH2 has a molecular weight of approximately 123 kDa. Scientists have identified its expression in various tissues though it is observed significantly in the brain testis and kidney. It participates actively in bridging components of the cytoskeleton and membrane signaling complexes.
Biological function summary

DIAPH2 influences cellular processes by regulating actin filament elongation and stabilization. This protein associates with the Rho family of GTPases forming a critical complex that drives localized actin assembly. This actin polymerization is necessary for processes like cell division cell migration and maintenance of cell shape. Additionally DIAPH2 plays roles in vesicle trafficking and the formation of cellular structures like filopodia and lamellipodia linking membrane processes with cytoskeletal elements.

Pathways

DIAPH2 integrates into significant pathways such as the Rho-GTPase pathway where it acts as an effector that transduces signals from upstream activators like Rho and Rac proteins. These pathways regulate the assembly and disassembly of actin filaments critical for various cell functions. Through its interactions with other proteins such as mDia1 and ROCK DIAPH2 influences the actin cytoskeleton impacting pathways involved in cell motility and adhesion.

Researchers have linked mutations in the DIAPH2 gene with disorders like premature ovarian failure (POF) and certain types of cancer. In POF DIAPH2 disrupts normal ovarian function impacting fertility. In cancer aberrant regulation of DIAPH2 can lead to improper cell division and tumor growth. Studies have also suggested interactions with proteins such as BRCA1 indicating a potential role in pathways involved in maintaining genomic stability.
