DIAPH3 also known as diaphanous-related formin 3 is a protein with an approximate mass of 130 kDa. It plays an important role in the regulation of the actin cytoskeleton essential for the dynamic reorganization of actin filaments. DIAPH3 primarily functions by mediating actin nucleation and elongation facilitating cellular processes such as shape changes adhesion and division. The protein is expressed in various tissues with notable expression in the brain testis and pancreas.
Biological function summary

DIAPH3 is a part of the formin family and acts by promoting the polymerization of actin filaments. It forms a complex with profilin and other actin-binding proteins maintaining the balance of cellular architecture. DIAPH3 contributes to the maintenance of normal cellular structure and dynamics participating in cell migration and morphogenesis. Its interaction with GTPases like RhoA regulates the initiation and stabilization of actin filaments directly impacting cell mechanics.

Actin nucleation and elongation factor required for the assembly of F-actin structures, such as actin cables and stress fibers. Required for cytokinesis, stress fiber formation and transcriptional activation of the serum response factor. Binds to GTP-bound form of Rho and to profilin : acts in a Rho-dependent manner to recruit profilin to the membrane, where it promotes actin polymerization. DFR proteins couple Rho and Src tyrosine kinase during signaling and the regulation of actin dynamics. Also acts as an actin nucleation and elongation factor in the nucleus by promoting nuclear actin polymerization inside the nucleus to drive serum-dependent SRF-MRTFA activity.
