Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease (By similarity).

Dickkopf-related protein 4 (DKK4) acts as an inhibitor of the Wnt signaling pathway which affects cell fate proliferation and migration. Known aliases for DKK4 include DKK-4 and DKK-4 protein. It possesses a molecular mass of around 27 kDa. DKK4 mainly expresses in the skin gastrointestinal tract and placenta indicating roles in these tissues and systems. Its presence suggests involvement in processes related to epithelial and stromal interactions.
DKK4 participates in the regulation of embryonic development and tissue homeostasis. It functions as a part of the larger Dickkopf protein family which modulates Wnt signaling by binding to LRP5/6 co-receptors. Through this interaction DKK4 manages the balance between cell growth and apoptosis important in both early development and adult tissue maintenance. DKK4 presence is often necessary to modulate signals that govern differentiation and proliferation in complex tissue systems.
DKK4 predominantly interacts within the Wnt signaling pathway. This pathway is critical for regulating gene expression involved in stem cell maintenance and differentiation. DKK4 acts by interacting with proteins such as LRP6 and Kremen thereby inhibiting Wnt-induced processes. Through modulation of these factors DKK4 ensures the fine-tuning of signaling cascades necessary for maintaining cellular equilibrium under physiological conditions.
Research associates DKK4 with malignant melanoma and colorectal cancer. These associations arise due to DKK4's role in modulating Wnt signaling which can drive oncogenic processes if dysregulated. In melanoma DKK4 can affect tumor cell proliferation by interacting with proteins like beta-catenin. In colorectal cancer abnormal expression of DKK4 might disrupt normal signaling balance further implicating it in tumor growth and progression. These connections highlight DKK4's potential as both a biomarker and a therapeutic target.
