Supplementary information

The protein Discs Large MAGUK Scaffold Protein 5 known as DLG5 functions as a scaffolding protein. It is part of the membrane-associated guanylate kinase (MAGUK) family and has an approximate mass of 220 kDa. DLG5 contains multiple domains including PDZ SH3 and GK which assist in protein-protein interactions. This protein is expressed in various tissues including the heart brain and skeletal muscle implying its widespread importance in different physiological contexts.
Biological function summary

The DLG5 protein plays a role in cell polarity and tissue development. It associates with components of the cytoskeleton and participates in maintaining the structural integrity of cells. DLG5 is part of a larger protein complex that regulates the signaling pathways important for cell differentiation and proliferation. Its interactions are essential for establishing cellular junctions that uphold cell adhesion and tissue architecture.

Pathways

The DLG5 protein is an integral part of the Hippo and Wnt signaling pathways. These pathways are critical in regulating cell growth and apoptosis. In the Wnt pathway DLG5 interacts with other proteins such as beta-catenin to influence gene transcription that steers cell cycle progression. In the Hippo pathway DLG5 interfaces with the LATS1/2 kinases to maintain control over organ size by modulating cell proliferation and apoptosis.

The DLG5 protein shows a connection to inflammatory bowel disease and some cancers. Mutations or altered expression of DLG5 have been linked to Crohn's disease suggesting its role in intestinal barrier function. Additionally its involvement in cancer highlights a relationship with tumor suppressor pathways. Through these diseases the protein interacts with others like APC and TCF within pathways that change cell adhesion and proliferation potentially assisting tumor development or impacting inflammatory responses.

Target data

Acts as a regulator of the Hippo signaling pathway (PubMed : 28087714, PubMed : 28169360). Negatively regulates the Hippo signaling pathway by mediating the interaction of MARK3 with STK3/4, bringing them together to promote MARK3-dependent hyperphosphorylation and inactivation of STK3 kinase activity toward LATS1 (PubMed : 28087714). Positively regulates the Hippo signaling pathway by mediating the interaction of SCRIB with STK4/MST1 and LATS1 which is important for the activation of the Hippo signaling pathway. Involved in regulating cell proliferation, maintenance of epithelial polarity, epithelial-mesenchymal transition (EMT), cell migration and invasion (PubMed : 28169360). Plays an important role in dendritic spine formation and synaptogenesis in cortical neurons; regulates synaptogenesis by enhancing the cell surface localization of N-cadherin. Acts as a positive regulator of hedgehog (Hh) signaling pathway. Plays a critical role in the early point of the SMO activity cycle by interacting with SMO at the ciliary base to induce the accumulation of KIF7 and GLI2 at the ciliary tip for GLI2 activation (By similarity).
