This antibody was developed through collaboration with the lab of Maria Morasso at the National Cancer Institute, NIH.

Activity summary

DLX3 also known as Distal-less homeobox 3 or BPES3 functions mechanically as a transcription factor with a molecular weight of approximately 30 kDa. This protein localizes mainly to the nucleus and is an essential regulator of gene expression. It is observed in tissues such as the epidermis placenta and developing teeth. By binding to specific DNA sequences DLX3 influences the transcriptional activity of various genes important for development.

Biological function summary

DLX3 participates in the regulation of cell differentiation processes during development. It acts individually and as part of a larger protein complex that influences the development of craniofacial structures and appendages by controlling gene expression in these areas. Its expression patterns suggest that it plays a vital role in morphogenesis and tissue specificity contributing to the development and compartmentalization within tissues.

Pathways

DLX3 interacts with key developmental signaling cascades notably the Wnt and BMP (Bone Morphogenetic Protein) pathways. These pathways regulate cellular differentiation and morphogenesis. In these pathways DLX3 indirectly interacts with proteins such as LEF1 and SMAD which are themselves involved in mediating the downstream effects of Wnt and BMP signaling. DLX3's role in these pathways highlights its importance in coordinating gene regulatory networks that drive organogenesis and development.

Associated diseases and disorders

DLX3 mutations are associated with Tricho-Dento-Osseous (TDO) syndrome and amelogenesis imperfecta affecting hair teeth and bone structure. DLX3 interacts with proteins like MSX1 and RUNX2 in the pathogenesis of these conditions which are characterized by defects in craniofacial and dental development. Understanding these interactions allows for a better comprehension of the molecular mechanisms underlying these disorders and potential therapeutic targets.

    DLX3 was immunoprecipitated from Mouse skin tissue lysate with Anti-DLX3 antibody [EPR8762(2)] ab178428 at 1/50 dilution (2μg in 0.35mg lysate). Western blot was performed on the immunoprecipitate using Anti-DLX3 antibody [EPR8762(2)] ab178428 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (VeriBlot for IP Detection Reagent (HRP) ab131366) was used at 1/1000 dilution.
